Name and Intended Use
The CHEMICOS Bovine Pregnancy Rapid Test is a rapid method for the detection of pregnancy-associated glycoproteins (PAGs) in plasma (EDTA) or serum of cows and heifers as a marker for pregnancy on farm.

General Information
Accurate and timely detection of pregnancy is an essential component of today's bovine reproductive management programs. Veterinarians and farmers use early detection of non-pregnant (open) cows and heifers to enable faster rebreeding and shorten the calving interval,
thereby maximizing new stock, milk production and revenue for the farm. Confirmation during the stages of gestation and at dry-off are also very economically important. In addition, beef producers may check for pregnancy at the many stages of production. Traditionally, pregnancy is determined through rectal palpation of the cow or by transrectal ultrasound (TU). The CHEMICOS Bovine Pregnancy Test provides users a method for accurate detection of pregnancy as early as 28 days post-breeding, and throughout gestation, which can give veterinarians, dairy farmers and beef producers an important tool for the early identification of pregnant cows. The test can be used ≥ 70 days post calving.

Product Principle
Pregnancy-associated glycoproteins (PAG) are specific proteins that appear in the peripheral blood of ruminant animals after pregnancy, playing a significant role throughout the pregnancy process. Secreted by binucleate cells of the embryo, they can be detected within 15-21 days of gestation in cows and have been applied for pregnancy diagnosis after 21 days of mating. In beef cattle research, the components of glycoproteins have also been detected significantly beyond 28 days. The Cow Early Pregnancy Colloidal Gold Rapid Test Card utilizes the principles of antigen-antibody specific binding in immunology, employing colloidal gold immunochromatography technology. Using a strip-like fibrous chromatographic material as the solid phase, the test rapidly determines the content of pregnancy-associated glycoprotein (PAG) in serum or plasma (EDTA) by facilitating sample solution migration through capillary action to a designated reaction and coloration area.

Components and Storage Conditions

(2) Storage Conditions for Test Cards:
The test cards should be stored in a cool, dry place at temperatures between 2°C and 25°C. Do not freeze. When stored properly, the reagents remain stable until the expiration date. Prior to experimentation, all components, including samples, must be equilibrated to temperatures between 15°C and 30°C.
Serum/Plasma samples should be stored at 2°C to 8°C and must not be stored for more than 24 hours.

  • Do not use expired test cards; use within the specified shelf life.
  • Do not touch or damage the white membrane in the center of the test card.
  • Do not mix test cards and dilution solutions from different batches.
  • Test cards are for one-time use; do not reuse.
  • Do not eat, drink, or smoke where samples or reagents are being handled.
  • Open the packaging before conducting the test. If the packaging is damaged or improperly sealed, do not use the pregnancy test card.
  • Do not expose the pregnancy test card to strong light before or during testing.
  • Do not conduct the test under direct sunlight.

Operating Instructions
  1. Before conducting the experiment, carefully read the operating instructions and allow both the test card and the test sample to reach room temperature.
  2. Blood Collection: Collect blood from the tail base of cows mated for ≥ 28 days. Lift the cow's tail and, approximately 10 centimeters from the tail base, insert the blood collection needle at the midpoint of the 3rd and 4th coccygeal vertebrae. Attach the other end to a centrifuge tube and draw 1-2 milliliters of blood (see Figure 1).
  3. Sample Extraction: Centrifuge at 4000 RPM for 10 minutes to separate serum or let it stand undisturbed for 2 hours to obtain serum (see Figure 2).
  4. Test Interpretation:
    • Serum/Plasma Sample: Remove the test card from its original packaging, mark it appropriately, and use a pipette to add 3 drops of upper-layer serum into the sample well (or use a pipette to add 90μl). Wait for 15 minutes to interpret the test result (see Figure 3).


Interpretation of Results

Positive:
Presence of a red or pink test line, T, indicates that the animal is Pregnant. The test line may range from dark to very light. The presence of a red or pink test line, T, at any intensity is considered Pregnant.

Negative:
Absence of test line indicates that the animal is Open. Retest after 7 days is recommended.

Invalid:
The test is invalid if the control line, C, does not appear. In this case, read the instructions carefully again and retest with a new test card.

Note: If the test cards need to be archived for record-keeping purposes, please air dry them after interpreting the results before storing.
